Mechanism & research

Retatrutide is a triple agonist targeting the GLP-1, GIP, and glucagon receptors. Through GLP-1 and GIP activation it enhances glucose-dependent insulin secretion and slows gastric emptying, while glucagon receptor agonism increases basal energy expenditure and hepatic lipid oxidation. In phase 2 clinical research, weekly dosing produced dose-dependent reductions in body weight averaging ~17–24% at 48 weeks, alongside improvements in fasting glucose, lipid profiles, and blood pressure.
