Mechanism & research

Melanotan I (afamelanotide) is a selective MC1R agonist that stimulates eumelanin synthesis in epidermal melanocytes, increasing UV-protective pigmentation. Melanotan II is a non-selective agonist at MC1R, MC3R, MC4R, and MC5R; central MC4R activation drives appetite suppression and pro-erectile signalling via dopaminergic pathways, while MC3R/MC5R activity influences lipolysis and sebaceous output. Together they produce darker, faster onset pigmentation alongside the libido and satiety effects unique to MC4R agonism.
